Metrobus launches the new service on Saturday (9 May) connecting Eastbourne, Uckfield and local villages with Gatwick Airport.
The 500 will also serve Hailsham, East Hoathly, Maresfield, Nutley, Forest Row and East Grinstead.
Buses will be operating 24/7 and will replace the existing services 54 between Eastbourne and Uckfield and the 261 between Uckfield and East Grinstead.
The maximum adult fare of £3 will apply to this route.
